The white spot being only on one side could also mean that you have been posting only on one diagonal, thus building up one shoulder more than the other.  If you do not change posting diagonals regularly, you'll end up with your horse having one muscled up shoulder and one much less muscled up, which can lead to the saddle pinching only one side.

As someone else has said, if there is a white spot and thickening around her withers, the saddle does NOT fit right. And the spot being on only one side indicates that maybe you are not riding centered, but putting more weight on one side.
