On a 6 hr trip, I'd probably just drive straight through.  On longer trips, we unload every 4-5 hours, depending on where good stopping places are, and offer water, let them graze, walk them around, etc.  Time out of the trailer is at least 15 minutes, sometimes 30 minutes if we're also eating lunch at the same time.  Letting them out gives them a chance to move around, lower their heads, loosen up their muscles, etc.  I know I always feel better after getting out of the truck and walking around a bit.  Even if they've already peed in the trailer, first thing they usually do upon getting out of the trailer is pee.
